I was all excited! I had met the staff of the Animal Emergency Treatment Center of Grayslake at the Family Pet Expo at Arlington Race track this past?March. They needed Dogs & Cats as blood donors for their blood bank.

What are the Requirements?

CATS:

*Greater than 10 pounds, 1-7 years of age, current on vaccinations and Feline Leukemia/Feline Immunodeficiency Virus negative, indoor only, in good health, no history of breeding or previous blood transfusions, and must have a good disposition.

My 4 year old cat Kaize is the ideal candidate for it! She is just as sweet & social as can be. ?She is 4 years old & in purrfect health! ?I figured it was a great way to ?Give Back? & help out other peoples pets. ?So I signed her up for the program.

At our first scheduled visit Kaize was to get a physical exam, blood typed & blood screen panel done. ?The technicican came into the exam room, explained how the screening process works, gave me a hand out on it with information & took Kaize in back. ?She returned about 10 minutes later with Kaize and told me she has Type ?A? blood. There is Type ?A? (Common), Type ?B? (less common) & Type ?AB? (rare). ?However, Kaize didn?t pass the physical exam portion because the technician told me they heard a 2/6 grade heart murmur. The only way she can continue the screen process & be allowed in the donor program is if she is screened & cleared by a cardiologist which I?d have to pay for.

I love my cats to death ? they are my kids. I?d do anything for them & only want the best for them. But thanks to my medical training I know that this news is nothing to panic or freak out about. ?I never saw or even met the veterinarian who supposedly heard the murmur. I?ve seen it first hand ?where one vet hears something & another doesn?t. I?m not about to run out & spend in upwards of $500+ to have her referred & seen by a cardiologist so she can be cleared to be a blood donor for a clinic that is 45 minutes away based on the opinion of a vet I never saw or met!

Kaize was just seen by her vet in April for her annual exam & vaccinations. She was given the all clear. So I made an appointment to have my vet see her & get their opinion. I?m willing to bet even if they do hear something they are going to just keep an eye on it & monitor it themselves. As she is only 4 years old & asymptomatic. If my vet recommends Kaize see a cardiologist I will make an appointment with one. So much for trying to do a good deed huh? LOL

So the following morning I?called AETC to find out of a report of last nights exam & findings will be sent to my vet. I was told they talked to the technician and a report was not written & nothing was typed up because it was found upon physical exam & the screening didn?t go any further. But they could give me the details over the phone. REALLY!? Free or not an animal is in your clinic, gets blood typed & has a physical exam but you don?t put in a report or type anything into your system about your findings?!

A heart murmur is an abnormal heart sound caused by the vibration of turbulent blood flow. Normally, blood flow is laminar and very quiet. Having a heart murmur is not itself an illness or disease. It is a characterization given to an abnormal sound heard upon auscultation over the region of the heart. Cats of any age, breed or gender can be born with or later develop heart murmurs. Many affected cats show no outward signs of having heart murmurs. In fact, most murmurs are detected incidentally during routine veterinary check-ups. When heart murmurs do accompany clinical signs, generally there is something more serious going on with the animal?s heart and the murmur is a consequence of ? not a cause of ? that underlying condition.

Murmurs are a clinical finding that cannot be ?prevented? or even ?treated? in the normal sense of those terms. The underlying cause of the murmur must be found. Many heart murmurs will never require medical or surgical management, even if they are very pronounced. Minor murmurs can often be managed with exercise and/or dietary change. Surgical procedures may be available for cats with more serious heart conditions causing their murmurs, although presently surgery is more commonly done in dogs.

I?m very interested to see what my veterinarian has to say about all of this when we see him Saturday July 7th!

Two years later?

I had my own office. I was a manager. Manager of half of the customer service representatives. I got a pay increase, health insurance, dental insurance, vision insurance, paid vacations, the whole working man?s gig. I had more money that I ever thought I?d have in my savings account. Shit, I had a savings account! I had enough money to buy a new car, a new TV, I got a cell phone, a good one, the kind that had the game Snake on it, a new sofa, and a new computer. Not that crappy thing Xavier gave to me years back, but one that was really fast, held a lot more memory, and I was able to afford DSL, a new thing to connect to and surf the internet faster.

I wore suits to work. Nice shoes, a crisp tie, pressed slacks, a form fitting blazer, and my hair was clean cut. Even when I got home from work, I didn?t immediately get into my pajamas like I used to. I?d loosen my tie, take off my jacket, and cook myself a good steak, some steamed vegetables, and drink a glass of wine. I gained twenty pounds.

I hung out with my coworkers a lot. We?d go to bars or restaurants after work, eat things that were more expensive that fast food hamburgers and sodas. We?d talk about the new company program implemented the week before, the new girl in accounting, or planned a weekend to Vegas.

Five days a week, woke up at 6 in the morning, got to work by 7:50, went to lunch at 12:35, left work at 3:30, met up with my coworkers somewhere, eat or drink, for a couple of hours, and then got home by 8pm. Watched TV for a couple more, fell asleep, got up at 6 and did the same thing. On the weekends, I?d meet up with coworkers and drink more, eat more, talk to girls more, and then wait for Monday to come around to start it all over again.

Two years, that was my life.?I lived comfortably. I was content. It was all a blur.

Researchers at the California Institute of Technology (Caltech) have been able, for the first time, to watch viruses infecting individual bacteria by transferring their DNA, and to measure the rate at which that transfer occurs. Shedding light on the early stages of infection by this type of virus?a bacteriophage?the scientists have determined that it is the cells targeted for infection, rather than the amount of genetic material within the viruses themselves, that dictate how quickly the bacteriophage's DNA is transferred.

"The beauty of our experiment is we were able to watch individual viruses infecting individual bacteria,"says Rob Phillips, the Fred and Nancy Morris Professor of Biophysics and Biology at Caltech and the principal investigator on the new study. "Other studies of the rate of infection have involved bulk measurements. With our methods, you can actually watch as a virus shoots out its DNA."

The new methods and results are described in a paper titled "A Single-Molecule Hershey?Chase Experiment," which will appear in the July 24 issue of the journal Current Biology and currently appears online. The lead authors of that paper, David Van Valen and David Wu, completed the work while graduate students in Phillips's group.

In the well-known 1952 Hershey-Chase experiment, Alfred Hershey and Martha Chase of the Carnegie Institution of Washington in Cold Spring Harbor convincingly confirmed earlier claims that DNA?and not protein?was the genetic material in cells. To prove this, the researchers used bacteriophages, which are able to infect bacteria using heads of tightly bundled DNA coated in a protein shell. Hershey and Chase radiolabeled sulfur, contained in the protein shell but not in the DNA, and phosphorous, found in the DNA but not in the protein shell. Then they let the bacteriophages infect the bacterial cells. When they isolated the cells and analyzed their contents, they found that only the radioactive phosphorous had made its way into the bacteria, proving that DNA is indeed the genetic material. The results also showed that, unlike the viruses that infect humans, bacteriophages transmit only their genetic information into their bacterial targets, leaving their "bodies" behind.

"This led, right from the get-go, to people wondering about the mechanism?about how the DNA gets out of the virus and into the infected cell," Phillips says. Several hypotheses have focused on the fact that the DNA in the virus is under a tremendous amount of pressure. Indeed, previous work has shown that the genetic material is under more pressure within its protein shell than champagne experiences in a corked bottle. After all, as Phillips says, "There are 16 microns [16,000 nanometers] of DNA in a tiny 50-nanometer-sized shell. It's like taking 500 meters of cable from the Golden Gate Bridge and putting it in the back of a FedEx truck."

Phillips's group wanted to find out whether that pressure plays a dominant role in transferring the DNA. Instead, he says, "What we discovered is that the thing that mattered most was not the pressure in the bacteriophage, but how much DNA was in the bacterial cell."

The researchers used a fluorescent dye to stain the DNA of two mutants of a bacteriophage known as lambda bacteriophage?one with a short genome and one with a longer genome?while that DNA was still inside the phage. Using a fluorescence microscope, they traced the glowing dye to see when and over what time period the viral DNA transferred from each phage into an E. coli bacterium. The mean ejection time was about five minutes, though that time varied considerably.

This was markedly different from what the group had seen previously when they ran a similar experiment in a test tube. In that earlier setup, they had essentially tricked the bacteriophages into ejecting their DNA into solution?a task that the phages completed in less than 10 seconds. In that case, once the phage with the longer genome had released enough DNA to make what remained inside the phage equal in length to the shorter genome, the two phages ejected DNA at the same rate. Therefore, Phillips's team reasoned, it was the amount of DNA in the phage that determined how quickly the DNA was transferred.

But Phillips says, "What was true in the test tube is not true in the cell." E. coli cells contain roughly 3 million proteins within a box that is roughly one micron (1,000 nanometers) on each side. Less than 10 nanometers separate each protein from its neighbors. "There's no room for anything else," Phillips says. "These cells are really crowded."

And so, when the bacteriophages try to inject their DNA into the cells, the factor that limits the rate of transfer is how jam-packed those cells are. "In this case," Phillips says, "it had more to do with the recipient, and less to do with the pressure that had built up inside the phage."

Looking toward the future, Phillips is interested in using the methods he and his team have developed to study different types of bacteriophages. He also wants to investigate various molecules that could be helping to actively pull the viral DNA into the cells. In the case of a bacteriophage called T7, for instance, previous work has shown that the host cell actually grabs onto the DNA and begins making copies of its genes before the virus has even delivered all the DNA into the cell. "We're curious whether that kind of mechanism is in play with the lambda bacteriophage," Phillips says.

The current findings have implications for the larger question of how biomolecules like DNA and proteins cross membranes in general, and not just into bacteria. Cells are full of membranes that divide them into separate compartments and that separate entire cells from the rest of the world. Much of the business of cellular life involves getting molecules across those barriers. "This process that we've been studying is one of the most elementary examples of what you could call polymer translocation or getting macromolecules across membranes," Phillips says. "We are starting to figure out the physics behind that process."

Afghanistan Violence: Insurgent Attacks Kill 10 Police Officers In Past 24 Hours

KABUL, Afghanistan (AP) ? Insurgent attacks in several parts of Afghanistan have killed 10 police officers over 24 hours, officials said Wednesday.

Warming weather in the summer usually brings an uptick in insurgent activity, but the surge this year is more than expected. Data from the NATO military coalition in Afghanistan show that attacks increased sharply in May compared with same time of year in 2011, from around 2,500 incidents to around 3,000. Statistics for June were not yet available.

In the south, four officers were killed when they responded to an explosion in Musa Qala district on Wednesday, said provincial spokesman Daoud Ahmadi. No one was killed in the first blast, but a second, apparently remote-controlled, bomb hit the policemen when they got to the scene to investigate.

In Herat province in the west, an ambush killed four policemen on patrol in Ghoryan district late Tuesday, said provincial government spokesman Noor Khan.

Also, in Kunduz province in the north, two police officers were killed when their truck hit a roadside bomb early Wednesday, said Kunduz province police chief Gen. Samihullah Qatrah.

'Wild Things' author spoke of desire to kill Bush

In one of his final interviews, celebrated children's book author Maurice Sendak said he fantasized about killing former President George W. Bush and Vice President Dick Cheney.

The "Where the Wild Things Are" author and illustrator, who died on May 8 at age 83, spoke with Gary Groth for an interview in The Comics Journal. A preview of the interview was published last month.

The famously cranky Sendak?spoke at length about his comics career,?his life and times, and blowing up the president.

"Bush was president, I thought, 'Be brave. Tie a bomb to your shirt. Insist on going to the White House,'" Sendak told Groth. "And I wanna have a big hug with the vice president, definitely. And his wife, and the president, and his wife, and anybody else that can fit into the love hug.

"And then we?ll blow ourselves up, and I?d be a hero," Sendak said. "To hell with the kiddie books. He killed Bush. He killed the vice president. Oh my God. ... It would have been a very brave and wonderful thing. But I didn?t do it; I didn?t do it."

Eric Reynolds, associate publisher at Fantagraphics Books, where Groth is founder and president, said Tuesday that they've seen quite a bit of outrage on Twitter and a few?conservative blogs. "I saw a tweet just a few minutes ago that said, 'I wish I'd read and collected Sendak's books as a kid so I could burn them now.'"

Sendak certainly never shied from expressing his outrage at numerous things. In an interview with The Guardian last fall, he railed against everything from e-books to the American political right to Gwyneth Paltrow.

"Anyone who has watched Sendak's infamous interview on the 'Colbert Report' (or read his books, for that matter), should recognize that not only was he cranky, he had a razor-sharp wit and a very dark sense of humor," Reynolds said. "He was 83 years old when he gave this interview [to Groth]. He was at the point in his life where he clearly didn't give a damn about propriety; he could speak his mind and clearly enjoyed provocation. I see these comments as part and parcel of his personality, not as a legitimate, actionable, treasonous threat."
